For the show's final season in 1965-66, the format was completely changed in order to capitalize on the popularity of spy shows like The Man from U.N.C.L.E. (1964) and Amos Burke himself became a secret agent. The title of the series was changed accordingly, but the new format proved unpopular and the show was cancelled.

This originated as the first installment of an anthology series, The Dick Powell Theatre: Who Killed Julie Greer? (1961). Dick Powell had played Inspector Burke on that show.

Amos Burke became a slightly different character in this series from the fellow he had been in "Who Killed Julie Greer", the opening episode of "The Dick Powell Show" from which this was spun off. Dick Powell, who had originally played him, died a few months before the series began to air, and Gene Barry, his replacement in the part, was some fifteen years younger. The character became much suaver, with emphasis on his designer clothes, his upper-crust social status and his playboy life-style. Also, Burke had now received a promotion in his police career, becoming a captain instead of an inspector.

Leon Lontoc, as Henry the chauffeur, was the only actor in "Who Killed Julie Greer" to become a series regular in "Burke's Law"; the character played by Regis Toomey in the series had been briefly featured in the original, but was there played by a different actor.
